83-5 利用葉綠素計量測菊花葉片之老化

利用葉綠素計量測菊花葉片之老化

摘  要
 

   本試驗之目的在建立以葉綠素計推估菊花葉綠素含量之方法,以應用於觀測切花老化過程中葉片黃化現象,並作為篩選品種之工具。葉綠素計量測之 SPAD 值與可萃取葉綠素含量之間,不論以葉面積或鮮重計算均有顯著相關。菊花在 25 ℃黑暗中誘導不同老化階段之葉片,其 SPAD 值與比色定量分析法之葉綠素含量為線性迴歸。以葉面積計算,其決定係數 R 2 =0.820 ,以葉鮮重計算,其決定係數 R 2 =0.660 。以具有不同深淺綠色葉片之 18 個品種, SPAD 讀值及葉綠素含量之線性迴歸分析,以葉面積及鮮重計算,其決定係數 R2 分別為 0.795 及 0.635 。觀察三個菊花品種葉圓片在黑暗中, 6 個不同溫度下之 SPAD 值變化,溫度愈高, SPAD 下降之速度愈快。測試了 18 個品種在 25 ℃黑暗中 SPAD 值變化。其 SPAD 初始值分別自 42.0 至 60.8 之間,培養 8 天後, SPAD 下降速度最快者為 ‘ 青心黃 ’ ,迴歸係數為 -6.51 ,下降速度最慢者為 ‘ 阿來粉 ’ ,迴歸係數為 -2.13 。高葉綠素含量之品種到達黃化 ( 約為 SPAD=30) 之日數較長 ( 相關係數 r=0.662) 。同時,葉片之黃化亦與葉綠素崩解之速率有關 ( 相關係數 r=0.718) 。在育種上,葉片葉綠素含量與其喪失速率二者均應做為選拔品種之指標。

Using Chlorophyll Meter to Investigate the Leaf Senescence of Chrysanthemum

ABSTRACT
 
   The aim of this study was to determine the relationship between extractable chlorophyll and SPAD readings in chrysanthemum leaves. Chlorophyll breakdown during leaf senescence was measured by chlorophyll meter, SPAD-502, and diversity of chlorophyll declination was evaluated among varieties.
  SPAD readings were significantly related to extracted chlorophyll of chrysanthemum leaves both on a fresh weight and leaf area basis. The linear regression relationship between the SPAD readings of leaves in different stage of senescence induced in 25 ℃ in the dark and chlorophyll contents analysed by spectrophotometry was described. T he R2 value for total chlorophyll on a leaf area basis was 0.820 , while on a fresh weight basis was 0.660. The SPAD readings of 18 varieties with different greenness and their extractable chlorophyll contents were also correlated. The R2 of linear regression equation on a leaf area basis was 0.795, while on a fresh weight basis was 0.635.
  Senescence of leaf discs of three varieties in 6 temperatures in the dark was investigated. The higher the temperatures, the faster the SPAD readings declined. Initial SPAD of 18 chrysanthemums were between 42.0 and 60.8. The speeds of chlorophyll declination in 25 ℃ in the dark among 18 varieties were calculated by regression analysis. The highest speed was SPAD –6.51 daily, and the lowest speed was –2.13. The estimated days to leaf yellowing of 18 varieties was correlated with the initial SPAD readings (r=0.662) and with declining speeds (r=0.718) of chlorophyll breakdown. It was suggested that chlorophyll content and its declination of leaf could be a selecting indicator of breeding program.
